Automated immunoassay kit for use on the Neptune instrument
This kit is intended to be performed on the Neptune Instrument. The assay is based on an enzyme immunoassay (EIA) principle. Each test strip consists of a membrane fixed onto a custom plastic support. During the fully automated procedure, the Neptune system sequentially incubates the test strips in ready-to-use reagent wells.
How it works:
The strip is first incubated with diluted patient serum. If specific human antibodies are present, they bind to corresponding antigens dotted on the membrane.
Unbound antibodies are removed via an automated wash step.
The strip is then incubated with alkaline phosphatase (AP)-conjugated goat anti-human IgG antibodies. These conjugates bind to the antibody-antigen complexes.
After a final wash, the strip is incubated in a chromogenic substrate solution.If enzyme activity is present, purple microdots appear on the membrane.
The signal intensity is directly proportional to the antibody concentration in the sample. Each test includes a 6-point calibration curve (including a blank) for fully quantitative analysis.

Multiple internal controls (sample, conjugate, and substrate) are included on every strip to ensure test validity from start to finish.

All dots are coated in triplicate, allowing for mean value calculation and confidence interval determination across antigens, controls, and calibration points.
